I believe Spencer is assuming the secret compound is Adcirca (aka Tadalafil). I'm not sure / do not recall whether that has been officially confirmed.

The transcript is available on SA if you are a paying member, otherwise you can search google for a cached copy. But IIRC it doesn't say anything more on the subject than what I quoted above.
